for me in the wrath of khan when kirk and saavik are talking in the lift then the door opens to McCoy saying "Whos holding up the damn elevator?" lol... simply great

I'm leaning more toward heroic or game-changing moments rather than just funny lines. If it was funny lines, we'd probably be here all day!
1. "I choose the danger...helluva time to ask" and taking on the rist of the Fal Tor Pan from Star Trek III
2. Figuring out how to trick the Vulcans with the "tri-ox" compound in Amok Time
3. Catching on to the "Vulcan Death Grip" ruse in The Enterprise Indident: "Your instincts are still good Mr. Spock! The captain is DEAD!"
4. Volunteering for the shuttle mission into the ameoba in The Immunity Syndrome (and later, "Shut up Spock...! We're rescuing you!")
5. His speech to Kirk in Balance of Terror "...don't destroy the one named Kirk."

His interchagne with Kahn as he has a knife to his throat in "Space Seed." Even in the face of death McCoy retains his spunk.
Many in the great and underrated "Journey to Babel." When he razzes Spock about having a "Teddy Bear" in his childhood, when he b tches and moans to Kirk about dressing up for the embassy, at the episode's end when he tells everyone to shut up so he can have the pleasure of getting the last word.
Mirror, Mirror- "I'm a Doctor, not an Engineer." Also, when he opted to save the "evil" Spock and risk his chance of escaping the Mirrorverse. Of course, this very decision led to Mirror Spock assisting them in getting back to their universe, thus making McCoy the hero of the episode.

Any one of his "I'm a Doctor, not a..." quotes.
Really, any episode in which he is fetured more prominently than Kirk or Spock.

That is hilarious I love that line too. I like a few minutes later he says "wonderful stuff that Romulan Ale"
I don't know if its really a "finest moment" but in Star Trek V when McCoy has to relive his pain was so emotional and Mr. Kelley really pulls off that scene so sad to watch him relive that.
Are you sure it isn't time for a colorful metaphor?

I agree, I think the scene in TFF was one of the most important and significant in the history of the character. It's ironic that DeForest Kelley originally protested the inclusion of this scene in the script. As it stands, it's one of my favorite McCoy scenes, and it's one of my favorite scenes in the whole movie franchise.

My ultimate favorite McCoy scene is this exchange between McCoy and Spock in the jail cell in "Bread and Circuses".
In the jail cell:
After McCoy tries to thank Spock for saving his life in the arena and Spock brushes him off, McCoy pushes Spock up against the jail wall and says:
"Do you know why you're not afraid to die, Spock? You're more afraid of living. Each day you stay alive is just one more day you might slip and let your human half peek out. That's it, isn't it? Why you wouldn't know what to do with a genuine, warm, decent feeling."
Spock replys, almost sadly: "Really, Doctor?"
McCoy says: "I know. I'm worried about Jim, too."
I've always appreciated this scene and have skipped through my dvd in the past just to review it. It's really great and does a wonderful job of displaying two important TOS facts: That McCoy really did like Spock all along despite his constant complaining, and that Spock was genuinely in emotional pain because of his breeding. Wonderful stuff.........

Every time he says: "I'm a Doctor, not a ~~~!"
or "He's dead, Jim."
And there are some pretty darn good ones from The Voyage Home, like before they leave Vulcan and he says "Angels and ministers of mercy, defend us."
Then: "He's not exactly working on all thruster, you know!"
And: (When they're at the PlexiGlass company and Scotty gets them a tour of the factory) "Don't bury yourself in the part!"
Dr. McCoy, simply put, is a comic genious sometimes......... one of my fave TOS characters.
